| Abstract | Antimicrobial resistance is an escalating global threat that demands a deeper understanding of bacterial responses to antibiotic stress. This chapter describes the application of label-free quantitative proteomics to investigate resistance- and antibiotic-induced changes in protein expression across clinically relevant Gram-positive and Gram-negative pathogens. Comparative proteomic analysis of methicillin-resistant and methicillin-sensitive Staphylococcus aureus identified protein expression changes associated with resistance and cellular adaptation. Proteomic profiling of a multidrug-resistant Escherichia coli strain treated with cefotaxime at the minimum inhibitory concentration revealed regulation of resistance-associated proteins and stress-response pathways linked to survival under β-lactam exposure. In Pseudomonas aeruginosa, proteome responses to tobramycin were examined in both clinical and reference strains, revealing strain-specific adaptive mechanisms. Additionally, mutant P. aeruginosa strains PvA and PvB were compared with the PAO1 reference strain to identify proteomic alterations associated with genetic variation. Collectively, these studies demonstrate the value of proteomics in elucidating mechanisms underlying antimicrobial resistance and antibiotic response. |
